![]() How to avoid cross-reactivity during immunohistochemistry/immunofluorescence experimentsĬross-reactivty might occur in immunostaining assays in tissue samples, for example while working is mouse tissues with mouse-derived antibodies. 10442-1-AP has a 71% homology with Sheep, and therefore a strong likelihood of reactivity. Step 3: Search for the organism (use the species' Latin name) you want to check homology with (under "Standard"). Step 2: Paste sequence into the NCBI-BLAST "query sequence" box. Step 1: Locate the immunogen sequence for the antibody (link to immunogen sequence can be found under "Product information"). Working example: What is the likelihood of Proteintech's P53 polyclonal antibody ( 10442-1-AP) cross-reacting in sheep samples (Ovis aries)? As mentioned above, you can use NCBI-BLAST and specifically select for your species of choice. It depends on the extent of protein sequence similarity between the immunogen and the potential cross-reactive protein sequence. Anything over 60% has a strong likelihood of cross-reacting, but will need to be verified in an assay and sample specific manner.ĭoes my antibody cross-react with other species? What is the “right number” for homology to predict cross-reactivity?īased on our extensive experience in antibody development and working with thousands of customers, the magic number for almost guaranteed cross-reactivity is 75% homology with the immunogen sequence. 95% of Proteintech's antibodies are raised against recombinant, long-length human proteins, meaning that they have an increased chance of cross-reactivity in non-human models such as mouse, rat, drosophila and zebrafish. Conversely, if you are looking for an antibody to work in other species, use a polyclonal antibody against a recombinant, long-length protein. Therefore if you suspect your antibody of cross-reacting negatively, consider switching to a monoclonal antibody (a homologous IgG antibody population only recognizing one epitope). Remember that polyclonal antibodies have a higher chance or cross-reactivity due to the fact that they recognise multiple epitopes along the immunogen sequence. Cross-reactivity is not always a negative quality, for example cross-reactivity of an antibody for a target across species allows the same antibody to be used in multiple model organisms. Here, you can check for an antibody's potential cross reactivity against that of similar proteins or in other species. This can be achieved by pair-wise sequence alignment using NCBI-BLAST. Thus, testing an antibody for cross-reactivity with closely related proteins is a to ensure accurate results.Ī quick and easy check for antibody cross reactivity is to assess the percentage homology of the antibody immunogen to that of other similar proteins. This is often the case when two antigens have similar structural regions that the antibody recognizes.Ĭross-reactivity can invalidate the results of an experiment and has negative impacts on scientific reproducibility. Cross-reactivity between antigens occurs when an antibody raised against one specific antigen has a competing high affinity toward a different antigen, therefore the antibody is able to recognize a protein which is different to the one it was raised against. An antibody has a specific amino acid sequence (the Fab region) that dictates its affinity for a specific antigen.

But I've also ripped my entire CD collection (~2,500 titles) to Redbook AIFF (using dBpoweramp). I do that for building playlists for iPod, background music, party tunes, etc. Its not really for critical listening but I would like for it to be the best it can be within reason. I just love the convenience of having my entire CD library at the tip of my fingers using the Remote app with an iPad. I have two choices from the Mac Mini to sound, both are via USB. One is to an Empirical Audio Offramp (with upgrades) and a digital coax out to preamp. The other, newest option, is USB straight to an asynchronous USB connection to the superb DAC section of a Rega Saturn-R. I honestly think I will only listen to these Redbook files and I don't think I'm ever really going to get on the hi-res train. For God's sake, I don't think I need another format to support. I think that might be enough background to finally get to my question. Before I try to untangle the mess of information regarding ALL of the "audiophile" players and make a choice between something like Amarra, PureMusic, jRiver, etc., I would like for someone to explain in terms that hopefully I can understand, what deleterious effect there is to just using iTunes by itself? Do these other programs just add features, that I may or may not have value for, or do they do something to make the music sound better? What? If this has been asked and answered someone point me in the right direction please. I've just recently tried the demo Audirvana, and am in the middle of a Pure Music trial. Testing was done with a grab-bag folder of 4000 tracks. They both sound slightly more robust than iTunes, and that in itself is dismaying. Seems to me any player should be sending the same 1s and 0s to the DAC. Pure Music's upsampling was very compelling, at first. But when I played familiar classics, it became evident it was mangling the files somehow. Worse, Pure Music is way too fiddly and finicky for my purposes. While they do visibly post instructions to minimize or disable many computer functions to optimize music playing, that's a non-starter for me. Might as well buy an outboard player in hardware form instead. Worst of all, PM occasionally stalls or stutters. (My iMac is loaded.)Īt least Audirvana is stable. Likely due to its independent-from-iTunes operation. I wouldn't call its library management great to work with, compared to iTunes. Guess I'll reluctantly buy this one only to play back the tiny number of DSD files in the system. Unless, as it's been suggested, it can't handle large libraries. I'm not sure what large is I don't think I'll ever exceed 50,000 tracks. My impression so far is that all these software players are tinker-toys compared to an outboard player. What is general opinion on Roon sound quality? I have used Audirvana+ for 5 years and updated each time a new version came out. I changed DACs 2 weeks ago, and my new DAC is more resolving of my server (Mac Mini with Uptone Audio MMK, LPS, SSD, 8 meg RAM feeding USB to my DAC. It sounds fantastic, but it got me thinking. So I spent all day on trying Amarra, HQ player, PureMusic, then lastly Roon. Audirvana+ is still ahead of all of these except Roon. I tried Roon about 18 months ago and it was nothing special. ![]() I don't upsample, and feed my NOS DAC resident rate. Swopping between A+ and Roon on select tracks, they sound very similar, closer than the rest of the bunch. But A+ has a digital sound to it, a very slight coarseness that clings to the treble region, most noticeable on female vocals for example. ![]() The controller system is made using the latest battery technology, Lithium Polymer, which has four times the life cycle of Lithium Ion but also absorbs energy more efficiently. The Great Santini became a hit film starring the widely respected Robert Duvall and Blythe Danner but not everything was roses during that particular period of his life. The story is one that delves deeper into a past that continued to plague his memories and it centered on his combined emotions of anger, frustration and love that he had for his dad. When 1976 rolled around, Conroy wrote the novel, “The Great Santini”, which would quickly become one of his greatest works. The Water is Wide had gone on to be turned into a film that went by the name, “Conrack” in 1974, starring Jon Voight. In 1972, the horrors that he saw would illuminate much of the theme of his very next novel, a memoir titled “The Water is Wide”. What he saw at the school was something that absolutely tore at his soul, which were conditions so deplorable and racism which ate at his heart. He not only wrote but published his very first book about his teacher whom he found to be his favorite teacher, “The Boo”.Īfter his graduation from the academy, Conroy had decided to become a teacher in an old fashioned, one-room schoolhouse. He began to attend the Citadel Military Academy in South Carolina and it was there when he had first decided to try his hand at writing. He would be at the receiving end of physical and verbal abuse but eventually he had drifted further away from his father by heading off to school. Born the first of seven children, Conroy had a truly uneasy upbringing because his father was an extremely abusive man who appeared to choose Pat as his daily whipping boy. The Prince of Tides went on to become made into a film that had garnered multiple Oscar nominations, starring Barbara Streisand and Nick Nolte.ĭuring the early years of Conroy’s life, he and his family moved around several different times due to military life and it wasn’t exactly easy when it came to the relationship of his father and him. Two particular novels of Pat Conroy that have become considered contemporary classics are “The Prince of Tides” and “The Great Santini”. The way his childhood and adolescence turned out, he clearly had a deep and lasting impression, one that would play a big role in many of his most-well known novels. When combining the FA values of efferent 1 with disease duration, the present DTI tractography analysis could be useful for differentiating MSA-C and CCA patients. The present DTI tractography newly showed that the FA values of two afferent fiber tracts showed significant declines in MSA-C patients, and afferent 1 showed good diagnostic sensitivity and specificity. Linear regressions showed strong correlations between FA value and disease duration in CCA patients (efferent 1, r = −0.466 afferent 2, r = −0.543 both p < 0.05), and between the FA value and the ratio of the standardized scale for the assessment and rating of ataxia (SARA)/disease duration in MSA-C patients (afferent 1, r = −0.407 p < 0.01). Receiver-operator characteristic curve analysis showed 85.7 % sensitivity and 75.0 % specificity of FA values in afferent 1 (cutoff value 0.476). Compared with CCA, MSA-C patients showed significant declines of fractional anisotropy (FA) values of afferent 1 and 2 ( p < 0.01, respectively) and a significant increase of the radial diffusivity (RD) value in afferent 1 ( p < 0.05). Tractography was performed using the DTI track module provided in the MedINRIA version 1.9.4, and regions of interest were drawn manually to reconstruct an efferent fiber tract and two afferent fiber tracts via the cerebellum. Forty-one MSA-C patients (62.7 ± 8.1 years old, mean ± SD) and age- and gender-matched 15 CCA patients (63.0 ± 8.6 years old) were examined. The objective of this study is to determine whether diffusion tensor imaging (DTI) tractography analysis is a potential method for differentiating cerebellar ataxia patients with multiple system atrophy with predominant cerebellar ataxia (MSA-C) and cortical cerebellar atrophy (CCA). Few games are able to showcase the power of the medium like Nier: Automata. If I had my way, every budding game developer would play this game to the point of acquiring endings A, B, C, D, and E before acquiescing to its final and alarmingly direct request. It is, as far as I’m concerned, historically significant in terms of its design and the way in which it uses the framework of a videogame to tell an evolving story. I will now post a quote from Jim Sterling's review that resonated with me so much that I cannot state it better as an ending to this lenghty review: "Nier itself is important. Yes, the fetch quests can be tedious and grinding can be a pain at times to achieve everything this game has to offer, but I did not waver and was fascinated by this world. From the amazing soundtrack by Keiichi Okabe to the wonderful directing and story by Yoko Taro and the combat by Platinum games.they all mesh together to create one of the best Action RPG's that I have ever played. This has been an eye opening experience that I will never forget. I won't go much into the story of the game, because it's very easy to spoil, but the story and overarching plot is so well done and it makes you question a lot of things about humanity, emotions, life itself and many other things. Imagine a Platinum game that has exquisite combat, a director that is so masterful in how he presents the story giving small information and tidbits as you go along and a soundrack by a composer that is well known (Keiichi Okabe) that you take a few moments from playing the game just to listen to the 's THAT GOOD. A game that shouldn't be passed on by ANYONE, because it does something that I, personally, haven't encountered in a game before (which I won't spoil here, because you HAVE TO experience it yourself). And what a fantastic job he and his team at Platinum Games have done. So without any doubt and after hearing that it got a PC release I instantly purchased it, because I was fascinated by what Yoko Taro could do. I will start by saying that I haven't played a Yoko Taro directed game before, but I have heard a lot of great things about his weird world that he has created that links between multiple games and franchises (Nier/Drakengard respectively) so I was really excited when I heard that Square decided to give him a good budget and a very well known studio for their character action games (Platinum) to make a sequel to the original Nier from the previous generation. Yoko Taro and his team have created a masterpiece that nobody should miss.
